Single Cell Gene Expression**

Single Cell Gene Expression provides users single cell transcriptome 3' gene expression in tens of thousands of cells. Scale experiments up or down with low-throughput and high-throughput solutions.

Single Cell Immune Profiling**

Single Cell Immune Profiling allows analysis of full-length, paired B-cell or T-cell receptor (BCR/TCR) sequences and 5’ gene expression, all from a single cell..

Fixed RNA profiling*

Fixed RNA Profiling enables single cell gene expression on precious samples that were previously inaccessible due to logistical challenges in sample handling. Fixation at the point of sample collection preserves fragile biology, reduce the batch effect and greatly streamlines workflows to provide critical insights on more samples.

No problem! We have a solution

Our expert comes to your laboratory, with the instruments and the equipments. We help you in the preparation of the cell suspension and we perform the first step of single cell suspension to ensure the optimal sample preparation. The RT-PCR amplification steps are also performed on site. Once the cDNA as a stable and easily transportable material is obtained, we carry it ourselves to GENXMAP to continue with the following steps of library preparation.

How does it work ?

Sample preparation :

The most critical step in ScRNA seq library preparation is single-cell suspension from your sample. If you are not using our on-site service, you can either cryopreserve or methanol-fix the cells and send your samples to our lab on dry ice, or if you are based closed to our lab (Marseille), it may be possible to use fresh single cell suspension. Please, contact us to discuss this option.

Sample processing, GEM generation and amplification :

Once we received your sample, we check the quality and count cells to ensure high quality of cell suspension. If necessary, we remove dead cells. Then we load the cell suspension on the chip according to 10x Genomics specifications, along with barcoded beads and reagents. The chip is placed in the machine to create GEM emulsion and to perform reverse transcription and barcoding. Once the 10x run is finished, we perform an amplification step to obtain cDNA.

Library preparation and sequencing :

We complete the NGS library preparation, we perform the QC on your library and the final step is to sequence your samples based on the specificity of your project by Nova Seq6000.

Parse Bioscience is a technology enables all scales of experiments without the limitations of today's microfluidics approaches. Combinatorial barcoding happens within cells themselves, resulting in extraordinary specificity and scalability. Split-pool combinatorial barcoding allows you to profile single-cell transcriptomes by assigning a specific barcode to each well into which cells are loaded (3-prime end capture method). We can take advantage of the cell itself as its own compartment to perform multiple rounds of splitting, pooling, and ligation of transcripts to generate different barcode combinations unique to each cell’s set of transcripts
